Sperm motility is estimated usually with a 4 grade system +++=progressive motility of more than >2.5 um/sec; ++ = weakly progressive motility - progessvie motility of less than 2.5 um/sec ; +=non-progressive motility - tail movement but no forward progression. 0= no tail movement visible................... One attempts to count 100 sperm recording their grading. Care needs to be taken to ensure that motile sperm are not overrepresented since motile sperm swin into your field of view whereas sperm with little or no motility do not. ONE Way to estimate the motility is to pause the video and count the sperm in the frame. Then run the frame and count the non-motile sperm. This will provide an estimate of motile over total sperm. Repeat for several frames and average to derieve a % motile. |
