IgG immunocoated beads will bind to sperm that contain antisperm antibodies bound to to their membrane. The test requries sufficient motile sperm and immunobead concentration to work. It may be performed either as a ditect test where ejaculated sperm already contain bound antbodies or as an indirect test where normal sperm are co-incubated with serum that may or may not contain antisperm antibodies. The sperm are allowed to co-incubate with serum for one hour and after washing, exposed to the immunobead. Binding may be either on the head, midpiece or tail are any combinbation. The number of sperm with immunobead binding are tallied out of 100 sperm counted and expressed as % of sperm with binding. |
